USAGE

Atazor is an anti retroviral drugs indicated for the treatment of HIV type I infection. Atazor helps to decrease the amount of HIV virus in blood. It lowers the chance of getting AIDS

PHARMACOLOGY

Atazor is a belongs to a protease inhibitor, this enzyme includes in cleavage of viral polyprotein Atazor is competent against HIV type I infection, it prohibits the cleavage of Gag-Pol polyprotein. This prevention occurs by Atazanavir active form of Atazor, link to protease enzyme and blocks the action produced by that appropriate enzyme So the prevention of cleavage causes to formation of immature cells.

DOSAGE

In adults:

HIV-1 infection

Treatment naive :
Atazor is concomitant use with other anti-retroviral medicine like, Ritonavir The usual dose is 300mg should be given orally as a single dose with Ritonavir 100mg If patient taking Ritonavir is not able to tolerate, 400mg of Atazor should be taken once a day with food While combination of Efavirenz with Atazor: 400mg PO with 100mg ritonavir qDay
Treatment experienced :
prescribed dose is 300mg orally as a single dose concomitant use with H2 receptor antagonist and Tenofovir is administeres 400mg PO with 100mg ritonavir qDay
Hepatic impairment :
For Mild hepatic impairment is given 400mg PO qDay For Moderate hepatic impairment is 300mg PO qDay Severe: not recommended
Renal impairment :
300mg PO with 100mg ritonavir qDay for Final stage of renal disease hemodialysis: In pediatric: 6-18 years (treatment naïve and treatment experienced) Atazor is not recommended for <15kg: 200mg PO with 100mg ritonavir qDay for 15kg to <35kg weighing patients 300mg PO with 100mg ritonavir qDay for ≥35kg ≥13 years (patient non tolerated to ritonavir) 400mg PO qDay for patient weighing ≥40kg

PHARMACOKINETIC

Absorption: The high serum concentration of the drug Atazanavir is occurred within 2.5 hours
Distribution: Highly bound to human plasma protein of Atazor (atazanavir) at nearly 86%. If Atazor taken with food, it may raises the bioavailability and decrease pharmacokinetic variability.
Metabolism :
In liver Atazor is largely metabolized and the two major biotransformation pathways in Atazor is Mono-oxygenation Di-oxygenation
Elimination :
The excretion exist via feces and urine. The Atazor half life of the drug is approximately 6 hours.

PRECAUTIONS

Atazor interaction with proton pump inhibitor should not used in treatment experienced patients. if patient getting severe rash or other anaphylactic reactions tablet Atazor should be stop Caution should be taken in case of Hepatic impairment Renal impairment High bilirubin levels

Atazor interaction with some of the list; Carbamazepine, lovastatin, Alfuzosin, astemizole, , Cisapride, Cobimetinib, dihydroergotamine, ivabradine, simvastatin, , pimozide, Phenobarbital, phenytoin, rifampin, sildenafil etc

CONTRAINDICATION

Patients are contraindicated to the ingredients present in the Atazor tablet Anaphylactic reaction occurs Atazor is contraindicated in the conditions; Chronic hepatitis B, C Diabetes Lactic acidosis Hemophilia Gallstones Kidney impairment High amount of bilirubin

MISSED DOSE

if patient fail to administer the dose, advice them to take within a time. Otherwise the missed dose should be skipped and follow the regular drug schedule
